Understanding extracellular electron transport of industrial microorganisms and optimization for production application (Frauke)

Abstract: Microbial electrosynthesis and electro-fermentation are novel approaches to increase microbial production by stimulating the cellular metabolism electrically. This thesis used a combined approach of in silico and in vivo strategies to understand the fundamentals of microbe-electrode interaction, the foundation and currently the limitation of the technology. Based on a metabolic network of Escherichia coli, several target processes for electro-fermentation were identified while simultaneously the effects of different electron transfer routes were modelled. Further, industrial relevant organisms were screened for their ability of extracellular electron uptake in vivo. The alcohol-producing acetogen, Clostridium autoethanogenum, was studied in-depth as a promising candidate for electrically-boosted production.

Title: Model-guided metabolic engineering of Saccharomyces cerevisiae for production of aromatics (Nils)

Abstract: Aromatics are amongst the most important feedstocks for the chemical industry. Petrochemistry-dependent production of these is increasingly becoming a matter of costs and sustainability, but to date no viable bioprocess exists. The shikimate pathway gives rise to a wealth of aromatic compounds that can replace current petroleum-based chemicals or be used for novel materials. In particular para-aminobenzoic (pABA), para-hydroxybenzoic (pHBA) and cis,cis-muconic acid, can be precursors for, but are not limited to, terephthalic or/and adipic acid, which are essential feedstocks for the production of PET and nylon. In this project a yeast-based microbial production system for aromatics was developed by combining metabolic modelling with genetic engineering.
